Ator, ativista. John nascido Charlton Carter, em 4 de outubro de 1923 (algumas fontes dizem 1924), em Evanston, Illinois, a mãe née Charlton e o pai Whitford Carter, um operador do moinho. Como único filho, cresceu numa área rural de St. Helen, Michigan. Com poucas crianças para fazer-lhe companhia, seu tempo de distração era ler livros. Seus pais divorciaram em 1930, e sua mãe casou mais tarde com Chester Heston. Charlton mudou-se com seus mãe e o padrasto para um suburbio perto de Chicago, onde estudou na High School. estudou depois na universidade do noroeste em 1942. Durante os dois anos seguintes, sua experiência teatral incluiu diversos papéis principais em produções dos estudante. Em 1944, deixou a faculdade e alistou-se no exército.
